There will be no real difference with different gauge wire in this case.

It is OK to run the laptop signal directly to the Loops.

Can I plug the guitar directly into the loops inputs or is it essential to go into the front gcx inputs as a ground for the guitar? I am thinking about getting a redeemer circuit in my guitar or a boost pedal like the lehle Sunday driver and wasn't sure if it would interact in a negative way with the front input buffers. I now plug into the front buffered guitar input and split the thru output to my tuner on my pedalboard. If I plug into the unbuffered input of the gcx or the loops input then I have no way to split the signal to my tuner. There lies my dilema but I could a splitter box if necessary.

Your guitar signal needs to be buffered in some way before the three way split in order to prevent completely loading down your pickups. Whichever method you prefer will be fine.
